Hi, 

I have been troubleshooting a nicoya openrev4xt for the past couple of months. I seem to be running into the issue of NSB, but am a bit stuck in ways I can combat this. I'm currently using a carboxyl sensor. 

My ligand is intrinsically disordered and has been shown to interact with BSA (past FP data), so I'm hesitant to introduce this. My analyte contains a zinc motif, which makes me nervous about using EDTA. 

What other options are there to combat NSB?

Hi,
Other additives that can be used are Casein or gelatin, but also here you should invesigate if your protein binds to it. Adding free carboxymethyl dextran mimics the sensor surface and may shield the binding moieties on your protein, add up to 1 mg/ml to the buffer.
Try a differen surface if possible, like a planar or ne coated with a hydrogel.
You are not mentioning the use of Tween-20 (use 0.005-0.1%).
