Hi
My lab will be acquiring a BIAcore X machine for a short period of time on a lease agreement and we were wondering about the requirements for setup (as we need to consider a limited budget):
A : Do we need to purchase all the acetate and borate buffers for preconcentration or can we make our own?
B : Does the BIAcore X machine require a pressurised gas canister?
C : Can we make our own regeneration buffers?
D : How many times can one use the NTA and Streptavidin chips?

A: it is possible to make your own. Use degassed water and high grade chemicals. Filter after prepearation. Store in aliqouts at -20C for long term.

B: I'am not familiar with the biacore X but i thought not.

C: Yes, just like A

D: this realy depends on what you do. NTA you can regenerate but with the SA-chip it is very hard to strip of bound proteins due to the nature of the SA-biotin binding.
