I am trying to set up SPR experiments with a protein that is stable only in Tris buffer. HEPES is definitely out of the question because it appears to bind to the protein and prevent it from binding to the appropriate ligand. I've noticed that biacore provides buffers with HEPES only. Could I possibly set up SPR experiments using Tris buffer?

Yes, all 'normal' buffer can be used in the Biacore.
Make sure you filter (0.22 um) and degas the buffer.
Also, you can not immobilize in TRIS buffer. Use PBS if possible.
