Considering streptavidin chips,

Similar to SPR platform, BLI (Octet, BioLayer Interferometry) also uses streptavidin coated biosensors. The immobilization pattern and assays are similar for SPR and BLI. They have given maximum/validated contact time of reagents with the BioLayer. Check Table.1 of the following document.

Also they mentioned that the higher contact time may damage the streptavidin surface. Can we possibly correlate with that data with Biacore SA chip stability?

Hi Charles,
This depends probably on the way that the streptavidin is immobilized to the sensor surface. For both BLI and SPR this is unknown. The BLI sensor is more flat than a SA-dextran chip in SPR thus possibly there are more SA molecules on a SPR sensor than on a BLI sensor. Shorter regeneration times are always better and we use 1-3 short pulses instead of one long one. Also a regeneration cocktail can be more effective at milder conditions ( www.sprpages.nl/kinetics/regeneration ).
